Pre-proposal

The goal of this program is to generate pilot data for a collaborative consortium manuscript using coral ancient DNA to reconstruct coral holobionts of the past. By recovering information from both the coral host and its associated microorganisms, including algal symbionts, bacteria, archaea, fungi, and viruses, we aim to examine how the composition and biology of the holobiont changed through time and across periods of extreme thermal stress. We are especially interested in well-dated samples for which the timing of thermal stress is independently supported by geochemical evidence from the coral skeleton or associated environmental records.

Each submitting group may propose three timepoints, ideally including one pre-industrial baseline and two timepoints spanning documented thermal stress events. We seek three biological replicates from each timepoint, for a total of nine samples and nine sequencing libraries per group. Applicants should briefly describe the rationale for the selected timepoints, the evidence supporting the timing and severity of the stress events, and the availability and suitability of the proposed biological replicates.
